AI-Designed Bacteriophages to Combat Drug-Resistant Bacteria (Source: The Hindu)

Context

Scientists have used AI-based genome language models to design complete genomes of bacteriophages, viruses that infect bacteria. Using bacteriophage ΦX174 (Phi X174) as a template, researchers produced functional synthetic phages capable of killing bacteria resistant to some naturally occurring viruses.

AI-Designed Bacteriophages: A New AMR Strategy

Explanation

  • Bacteriophages (phages) are viruses that specifically infect bacterial cells and are generally composed of genetic material (DNA or RNA) enclosed within a protein capsid.
  • During the lytic cycle, a phage attaches to a bacterium, injects its genome, replicates using the host machinery and causes cell lysis, releasing new phages.
  • Lysogenic phages can integrate their genetic material into the bacterial chromosome as a prophage without immediately killing the host.
  • Phage therapy uses bacteriophages to target pathogenic bacteria and is being explored as an alternative or complement to antibiotics, particularly against antimicrobial resistance (AMR).
  • Genome language models learn patterns in biological sequences and can generate novel DNA sequences; designing entire functional viral genomes represents an application of AI in synthetic biology.
  • ΦX174 is a well-studied bacteriophage with a small single-stranded DNA (ssDNA) genome, making it useful in molecular biology and synthetic-genome research.
